Community Update: Advancing Research Together

For YWHAG, every step forward is made possible by families who choose to participate in research efforts and by supporters who help fund the work that drives discovery. From contributing to studies, to sharing experiences, to supporting fundraisers, each family plays a role in accelerating progress.

Because YWHAG is ultra-rare, every data point matters. Every participant helps researchers better understand the condition, and every dollar raised helps fuel the studies that bring us closer to meaningful treatments.

Together, these efforts create momentum—and that momentum is what moves science forward.

During our latest community meeting, we were joined by leading researchers who shared insights into ongoing drug repurposing efforts and the role of our Natural History Study in shaping the path forward.

Progress in Drug Repurposing

Lovastatin was originally identified through the foundation’s drug repurposing efforts, where early laboratory studies suggested it may help normalize disrupted cellular signaling in YWHAG. While this work is still in the early stages, it represents a meaningful step toward identifying treatments that could make a difference for our children.

Importantly, this is not yet a formal clinical trial but it is part of a strategic process to better understand what may help.

The Role of the Observational Study

A major focus of the discussion was the importance of our observational study within the Natural History Study.

This effort allows families and clinicians to share real-world experiences, including: Changes in seizure activity, developmental progress, and behavioral observations

By gathering this information, we can begin to identify patterns that may help guide future clinical studies and research priorities.

This is especially critical in a rare condition like YWHAG, where every data point helps build a clearer picture.

Why Community Participation Matters

Progress in rare disease research depends on community involvement.

The observational study does not involve prescribing medications or changing care. Instead, it is a way to safely and responsibly learn from real-world experiences and ensure that future research is grounded in what families are actually seeing.

Every family that participates helps move the science forward—not just for today, but for what comes next.

Interested in Participating?

If you are currently using, considering, or simply interested in contributing to this observational effort, we would love to hear from you.

Supporting Conversations with Your Care Team

We recognize that navigating potential treatment options can feel overwhelming, especially when discussing emerging research with your child’s care team.

To help support families in these conversations, the Foundation has developed a clinician-friendly handout that summarizes:

  • The rationale behind drug repurposing in YWHAG

  • Why lovastatin has emerged as a potential candidate

  • Evidence from other rare and neurodevelopmental disorders where it has been safely and effectively used

  • Key considerations for discussion with healthcare providers

This resource is designed to be informative and easy to share with primary care physicians and specialists. It is not intended to replace medical guidance, but rather to help facilitate more informed and productive conversations between families and their care teams.

If you would like a copy of this handout, please reach out to us—we are happy to provide it.

We will continue to provide updates as we learn more and as this effort evolves. Together, we are building the foundation for future treatments and, ultimately, a cure.
